Les carcinomes nasopharyngés (NPC) présentent une surexpression constitutive de la protéine c-IAP2. A l'aide d'un inhibiteur pharmacologique de c-IAP2 et d'ARN interférents, j'ai montré qu'elle est essentielle pour la survie et la croissance clonogénique des cellules de NPC. Dans ces cellules, une fonction spécifique de c- IAP2 consiste ŕ bloquer la composante pro-apoptotique des signaux liés ŕ la stimulation du TLR-3. Une partie de ces résultats ont été étendus ŕ d'autres types de cellules malignes oů l'expression de base de c-IAP2 est faible mais augmente fortement sous l'effet d'une stimulation du TLR3. Cette augmentation passe par une activation du gčne BIRC3 codant c-IAP2. Lorsque la stimulation du TLR3 est combinée ŕ une inhibition de c-IAP2, il en résulte, dans une majorité de lignées cellulaires, une apoptose massive impliquant la caspase-8. Cette combinaison pourrait constituer la base de nouvelles approches thérapeutiques contre divers catégories de tumeurs malignes humaines.
